Briefly, mice were placed on a 10 × 20 cm screen (grid size 0.2 × 0.2 cm) that could be rotated from 0° (horizontal) to 90° (vertical). The mouse was placed on the horizontal screen, and the screen was then rotated into the vertical plane. The duration of time that the mouse was able to hold on to the vertical screen was recorded for a maximum of 15 seconds (allowing a total of 3 points). Then, the mouse was placed at the center of a horizontal wooden rod (1.5 cm in diameter), and the time that the mouse was able to remain balanced on the rod was recorded for a maximum of 30 seconds (allowing a total of 3 points). Finally, a prehensile traction test was administered. The time that the mouse was able to cling to a horizontal rope was recorded for a maximum of 5 seconds. From these tests, a total motor score (TMS) (9 possible points) was computed. The neurological tests were determined by an observer who was unaware of the grouping. The TMSs that we applied in this study have been proven to be a more accurate method to evaluate global cerebral ischemia in mice.

2.6. TUNEL staining
For detection of in situ DNA fragmentation, terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL) staining was performed using an In Situ Cell Death Detection Kit (Roche Diagnostics, Mannheim, Germany) according to the manufacturer’s instructions as described in a previous study (Ma et al., 2015). The total numbers of TUNEL positive neurons in the CA1 region were counted in 3 different fields for each section in a blind manner by fluorescence microscopy at ×400 magnification (BX-60; Olympus, Tokyo, Japan).The numbers of TUNEL-staining positive cells were recorded, and the differences were analyzed among the groups.

2.7. Western blot analysis
Hippocampus samples were used for mitochondrial isolation according to the manufacturer’s specifications (Qproteome mitochondria isolation kit, Qiagen, Hilden, Germany). The whole protein concentration was determined by the Bradford method, and Western blot was performed as previously described (Ma et al., 2015). The following primary antibodies were used: anti-cleaved caspase-3 polyclonal antibody (1:50), anti-Cyto C monoclonal antibody (1:5000), anti-AIF polyclonal antibody (1:1000), anti-COX IV polyclonal antibody (1:100), anti-histone polyclonal antibody (1:1000), anti-SOD2 polyclonal antibody (1:5000) and anti-β-actin polyclonal antibody (1:1000). Appropriate secondary horseradish peroxidase-conjugated goat anti-rabbit or goat anti-mouse antibodies (1:10000, CWBIO, Beijing, China) were used. Antigens were detected using the chemiluminescence technique (Amersham Pharmacia Biotech Piscataway, NJ, USA). Image analysis was accomplished with the assistance of computerized analysis software (Bio-Rad Laboratories, Hercules, CA).

2.8. Cell viability and lactate dehydrogenase (LDH) release measurements
The neurons were cultured in 96-well plates at a density of 1 × 105 cells/well. By using the MTT method, the cell viability was tested. Briefly, after the treatments, 20 µl MTT solution (5 mg/ml) was added into each well (96-well cell culture plate). After 4-h incubation at 37 °C, the medium in the well was removed and 150 µl DMSO was added into each well. After the formazan in the wells was dissolved completely, the absorbance was measured at a wavelength of 490 nm by using a spectrophotometer (TECAN, CH). The neurons were cultured in 24-well plates at a density of 2 × 105 cells/well. After the treatments, the supernatant of each well was removed to test the LDH activity, which was measured according to the manufacturer’s instructions of the LDH Reagent kit, which we described in one previous study (Jia et al., 2014).

2.9. Cell apoptotic rate assay
The apoptotic rate of treated neurons was tested by using a flow cytometry (BD, USA). Briefly, the cells were cultured in a 6-well plate at a density of 5 × 105 cells/well. After the treatments, the cells were harvested by centrifugation at 1000 rpm for 5 min. After two washes with ice-cold phosphate buffered saline (PBS), the cells were re-suspended in binding buffer at a density of 1 × 106 cells/ml. Next, 5 µl of fluorescein 5-isothiocyanate [2-(3,6-dihydroxy-9H-xanthen-9-yl)-5-isothiocyanatobenzoic acid] FITC-conjugated anti-annexin-V staining antibody and 2 µl of propidium iodide (PI) solution were added into 100 µl of the binding buffer. After mixing completely, the cells were incubated for 15 min at room temperature in darkness, and the apoptotic rate was assessed.

2.10. Mitochondria isolation and the extractions of cytosolic and nuclear protein
Mice were anesthetized by using 3 % isoflurane, decapitated and the brains were rapidly removed. The hippocampus tissue was removed, and the mitochondria were isolated from the tissue. Mitochondria, cytosolic protein and nuclear protein were isolated according to the manufacturer’s specifications (Qproteome mitochondria isolation kit, Qiagen, Hilden, Germany).

2.11. Transmission electron microscopy
Mice were anesthetized and transcardially perfused with normal saline and fixative composed of 4 % paraformaldehyde and 2.5 % glutaraldehyde. The hippocampal CA1 regions were removed for postfix with 3 % glutaraldehyde (pH 7.4, 4°C) and then fixed again in 1 % osmium tetroxide, dehydrated in graded ethanol, embedded in epon. 50-µm-thick hippocampal sections were cut with amicrotome (Bromma,Sweden), counterstained with uranyl acetate and lead citrate, and observed with a JEM-2000EX transmission electron microscope (JEOL, Japan).

2.12. Mitochondrial complex activity and mitochondrial membrane potential measurements

Mitochondrial complex I and IV activities were measured at 30 °C spectrophotometrically by the methods of a previous study (Han et al., 2007). Mitochondrial membrane potential (MMP) was determined by using JC-1 (Sigma-Aldrich, St. Louis, MO, USA). Mitochondria samples (0.5 mg/ml, 1 ml) were incubated with 19 ml JC-1 staining buffer according to the manufacture’s instruction. At the end of the experiments, valinomycin was added as a negative control. Fluorescence intensity was determined at 37 °C by using a fluorescence spectrophotometer (TECAN, CH). The ratio of aggregates (red, 590 nm) to monomer (green, 525 nm) was calculated as an indicator of MMP.

2.13. Measurement of mitochondrial swelling degree
Ca2+-induced mitochondrial swelling as measured by A520 was used as an indication of MPTP opening as previously described (Sullivan et al., 1999). Briefly, 1 mg of isolated mitochondria was suspended in 1 ml of respiration buffer plus 10 mM succinate. . The absorbance at 520 nm (A520) was measured with a spectrophotometer (TECAN, CH) at 36°C and a change of absorbance within 10 min indicated mitochondrial swelling. The difference (ΔA520) between the maximum A520 reading (immediately after CaCl2 addition) and the minimum A520 reading (10 min after CaCl2 addition) were used for statistical analysis.

2.14. Mitochondrial superoxide assay
MitoSOXTM Red reagent was taken to evaluate mitochondrial superoxide in this study. The mitochondrial superoxide level was tested as we described in a previous study (Jia et al., 2016). Briefly, MitoSOX™ targets the mitochondria selectively after permeating cell membrane and reacting with the mitochondrial superoxide rapidly.After the treatments, the hippocampal neurons received 20-min incubation with 5 µM MitoSOX™ Red at 37 °C. A confocal microscope (Olympus, Japan) was used to take the fluorescence photographs, including the mitochondrial superoxide (red, 510 nm at excitation and 580 nm at emission), the nuclei (blue, 340 nm at excitation and 488 nm at emission) and the merged photos. The fluorescence intensities of the mitochondrial superoxide photographs were measured by using image pro-plus software (IPP 6.0, Media Cybernetics, Silver Spring, MD, USA).

2.15. Statistical analysis
SPSS 13.0 for Windows (SPSS Inc., Chicago, IL) was used to conduct statistical analysis. Except for neurological scores, the other values were expressed as means ± SD, and were analyzed by one-way ANOVA. Between-group differences were detected with post hoc Student-Newman-Keuls tests. The neurological scores were expressed as the median, and were analyzed with Kruskal-Wallis test followed by the Mann-Whitney U test with Bonferroni correction. P < 0.05 indicated statistical significance. 3. Results
3.1. Cannabinoid CB1 receptor agonist ACEA induced neuroprotection against brain ischemia, and MPTP opener reversed the effects
To investigate the neuroprotective role of cannabinoid CB1 receptor in Blasticidin S datasheet brain ischemia, we took mouse BCCAO model to mimic the brain ischemia/reperfusion (I/R) injury. Compared with the sham group (Figure 1A), we found that 20-min brain ischemia reduced the total motor scores at 24, 48 and 72 h after the reperfusion significantly (P < 0.001), and administration of cannabinoid CB1 receptor agonist ACEA markedly increased the total motor scores compared with the brain ischemia (BCCAO) group (P = 0.016, 0.002 and 0.002 respectively). However, the use of MPTP opener atractyloside (Atr) obviously abolished the ACEA-induced ameliorations of total motor scores (P = 0.041, 0.007 and 0.005 respectively), compared to the ACEA+BCCAO group. And the MPTP inhibitor CsA did not cause significant effects on the ACEA-induced ameliorations of mice motor ability (P = 0.132, 0.093 and 0.163 respectively). Additionally, at 72 h after the I/R injury, we tested the ACEA-induced effects on apoptosis in ischemic hippocampus tissue. Similarly, compared with the sham group, we found that 20-min brain ischemia increased the number of TUNEL-staining positive neurons (P < 0.001, Figure 1B,C) and the cleaved caspase-3 (Cl. Cas-3) protein expression (P < 0.001, Figure 1D), and the ACEA diminished the TUNEL-staining positive neurons (P < 0.001) and reduced the cleaved caspase-3 protein expression (P < 0.001). The MPTP inhibitor Atr, significantly abolished the ACEA-induced effects on the TUNEL-staining positive cells counting (P < 0.001) and the cleaved caspase-3 protein expression (P = 0.002), however, the MPTP inhibitor CsA did not change the effects of ACEA (P = 0.598 and 0.827 respectively). These findings showed that inhibition of MPTP opening may be involved in CB1 receptor agonist ACEA-induced neuroprotection against brain ischemic injury.
3.2. Cannabinoid CB1 receptor agonist ACEA protected neurons against OGD injury, and MPTP opener reversed the protections
To further determine the CB1 receptor agonist ACEA-induced neuroprotection in vitro, we used primary cultured hippocampal neurons exposed to oxygen-glucose deprivation (OGD) to imitate the ischemic neuron. We used MTT to assess the cell viability, LDH reagent kits to evaluate the LDH concentrations in the supernatants and flow cytometry to measure the apoptotic rate of the cells (Figure 2A,D). At 24 h after re-oxygenation, the cell viability of the OGD group decreased, the LDH level and apoptotic rate increased significantly (P < 0.001), compared with the control group. Administration of ACEA significantly maintained the cell viability, reduced the LDH release and down-regulated the apoptotic rate (P < 0.001). The MPTP opener Atr obviously abolished the CB1 receptor agonist ACEA-induced neuroprotection above (P < 0.001), however, the MPTP inhibitor CsA did not change the effects of ACEA (P = 0.702, 0.415 and 0.454 respectively), suggesting that inhibition of the MPTP opening may be involved in the ACEA-induced neuroprotection against OGD in vitro. 3.3. The used doses of MPTP opener Atr and inhibitor CsA did not induce toxicity to the neuronal cells
To test whether the administrations of MPTP opener Atr and MPTP inhibitor CsA induce toxicological effects to neuronal cells in this study, we used total motor score to evaluate the neurological function and western blot to measure the apoptosis-associated cleaved caspase-3 protein expression in vivo, and took MTT and LDH release to assess the cell injury level in vitro. In vivo, the Atr of 5 mg/kg or CsA of 10 mg/kg was injected intracerebralventricularly. At 72 h after the injection, the Atr or CsA did not induce significant effects on the neurological function (P = 0.163 and 0.202 respectively) and cleaved caspase-3 protein expression (P = 0.155 and 0.925 respectively), compared with the vehicle group (Figure 3A,B). In vitro, the cells were exposed to the medium containing 50 µM Atr or 10 µM CsA for 24 h (Figure 3C,D). The MTT and LDH release analysis showed that Atr or CsA exposures did not reduce the cell viability (P = 0.358 and 0.520 respectively) or increase the LDH release (P = 0.082 and 0.372 respectively). These findings showed that the doses of Atr or CsA used in this study did not induce neurotoxicity to neuronal cells in vivo and in vitro.

3.4. Cannabinoid CB1 receptor agonist ACEA decreased the releases of cytochrome C and apoptosis-inducing factor from mitochondria in neurons exposed to OGD, and MPTP opener reversed the effects After brain ischemia, the mitochondrial permeability transition pore (MPTP) may open in the ischemic brain tissue. Following the MPTP opening, the cytochrome C (Cyto C) and apoptosis-inducing factor (AIF) can be released from the mitochondria into the cytosol and cellular nucleus respectively. Therefore, evaluating the MPTP opening level can be via testing the Cyto C level in cytosol and AIF level in cellular nucleus. In this experiment, we found that the Cyto C level in the cytosol was increased at 24 h after the OGD plus re-oxygenation (P < 0.001), compared to the control group (Figure 4A). And CB1 receptor agonist ACEA significantly reduced the Cyto C protein level in the cytosol (P < 0.001). However, the MPTP opener Atr obviously abolished the ACEA-induced down-regulation of the cytosolic Cyto C protein level (P = 0.002), and the MPTP inhibitor CsA did not cause significant effects on the ACEA-induced down-regulation of Cyto C level (P = 0.706). In contrast, the mitochondrial Cyto C protein level was reduced significantly at 24 h after the re-oxygenation, compared with the control group (P = 0.003). And ACEA maintained the mitochondrial Cyto C protein level (P = 0.01). However, the Atr (P = 0.01), not the CsA (P = 0.499), markedly reversed the ACEA-induced effects on the mitochondrial Cyto C level (Figure 4B).In addition, the total intracellular Cyto C expression did not change among the groups (P > 0.05, Figure 4C).
Similarly, the AIF protein level was increased in the neuronal nucleus and decreased in the mitochondria (Figure 4D,E) at 24 h after re-oxygenation, compared with the control group (P < 0.001). And the ACEA significantly inhibited the OGD plus re-oxygenation-induced AIF release from the mitochondria to the nucleus (P < 0.001). However, the MPTP opener Atr (P = 0.001 and 0.005 respectively), but not the MPTP inhibitor CsA (P = 0.458 and 0.486 respectively), obviously abolished the ACEA-induced AIF level changes above. And the total intracellular AIF expression was unchanged among the groups (P > 0.05, Figure 4F). These findings indicated that CB1 receptor agonist ACEA can inhibit the opening of MPTP in neurons treated with OGD plus re-oxygenation injury.

3.5. Cannabinoid CB1 receptor agonist inhibited mitochondrial swelling, maintained mitochondrial morphology and mitochondrial membrane potential in neuronal cells after the ischemia and OGD, and MPTP opener reversed the effects To further observe the opening neuronal mitochondria, we used electron microscope and specific reagent kits to study the mitochondrial morphology, degree of mitochondrial swelling and mitochondrial membrane potential (MMP). At 72 h after the I/R injury, morphology of the neuronal mitochondria (indicated by the black arrows, Figure 5A) enlarged obviously, and the mitochondrial outer and inner membranes were hardly distinguishable, compared to the elongated and easily distinguishable outer and inner membranes of mitochondria in the sham group.And the ACEA administration markedly reduced the mitochondrial enlargement and maintained the mitochondrial morphology. However, the MPTP opener Atr, not the MPTP inhibitor CsA, obviously abolished the ACEA-induced
morphological amelioration of mitochondria.After the treatments, the mitochondria suspensions were collected from the treated neurons. At 24 h after re-oxygenation, the mitochondrial swelling degree was increased dramatically compared with the control group (P < 0.001, Figure 5B), meanwhile, the mitochondrial membrane potential (MMP) level (Figure 5C) was decreased obviously (P < 0.001), and the CB1 receptor agonist ACEA significantly inhibited the mitochondrial swelling (P < 0.001) and maintained the MMP level (P = 0.005). However, the MPTP opener Atr (P = 0.001 and 0.003 respectively), but not the inhibitor CsA (P = 0.865 and 0.342 respectively), markedly reversed the ACEA-induced effects on the mitochondrial swelling and MMP level. 3.6. Cannabinoid CB1 receptor agonist reduced mitochondrial superoxide level, increased SOD2 expreesion and ameliorated mitochondrial complexes activities in OGD-treated neurons, and MPTP opener abolished the effects After the OGD and re-oxygenation treatment, the treated neurons may be injured by the oxidative stress. Therefore, we tested the mitochondrial superoxide level and the anti-oxidant SOD2 expression in mitochondria, meanwhile, we also measured the mitochondrial complex I and IV activities to know the mitochondria injury degree at 24 h after re-oxygenation in neurons. We found that the mitochondrial superoxide level was elevated (Figure 6A,B) and mitochondrial SOD2 expression was decreased (Figure 6C) in the OGD group (P < 0.001), compared to the control, and CB1 receptor agonist ACEA obviously reduced the mitochondrial superoxide level and increased the SOD2 expression (P < 0.001). Nevertheless, the MPTP opener Atr (P < 0.001 and P = 0.005 respectively), but not the inhibitor CsA (P = 0.932 and 0.775 respectively), reversed the ACEA-induced effects above.Similarly, the OGD treatment reduced the mitochondrial complex I and IV activities (P < 0.001, Figure 6D,E),and the ACEA exposure maintained the two mitochondrial complexes activities (P < 0.001 and P = 0.004 respectively). However, the MPTP opener Atr (r = 0.003 and 0.001 respectively), but not the CsA (r = 0.743 and 0.650 respectively), reversed the ACEA-induced effects on mitochondrial complex I and IV activities. These findings above indicated that the ACEA-induced anti-oxidation and protections in mitochondria may be via inhibiting the opening of MPTP. 4. Discussion
In this investigation, we found that cannabinoid CB1 receptor agonist ACEA significantly inhibited the brain ischemia-induced neurological dysfunction, neuronal cell apoptosis and mitochondrial morphology deterioration in brain tissue, reduced OGD-induced cell injury, mitochondrial Cyto C and AIF releases, mitochondrial swelling, mitochondrial superoxide accumulation and the consumption of mitochondrial SOD2, maintained the mitochondrial functions, leading to neuroprotection ultimately. However, the MPTP opener Atr obviously reversed the ACEA-induced neuroprotective effects, inhibited the mitochondrial Cyto C and AIF releases and relieved the mitochondrial swelling, but the MPTP inhibitor CsA did not cause significant effects on the ACEA-induced neuroprotection. These results indicated that inhibition of MPTP opening might participate in the ACEA-induced neuroprotective effects (Fig. 7). Neurons are extremely sensitive to ischemia and the other insults (Kislin et al., 2017), moreover, neuroregeneration is a tough work in the injured brain tissue (Cheng et al., 2017). For this reason, finding effective drugs and therapies for the treatment of brain ischemic injury, such as ischemic stroke, is of great importance. Some studies showed that cannabinoids can bring about neuroprotective effects against brain ischemic injury by activating cannabinoid (CB) receptors expressed in the brain tissue. There are two main cannabinoid receptors, CB1 and CB2 (Lopez-Rodriguez et al., 2015).

In the central nervous system (CNS), CB1 receptor is mainly expressed in neurons and astrocytes, and CB2 receptor is expressed in microglial cells and astrocytes (Stella, 2010; Maccarone et al., 2016). In neurons, CB1 receptors have been found in cell membrane and mitochondria (Bénard et al., 2012;Stornaiuolo et al., 2015), and no more evidences show that CB1 receptor is expressed in the other cell organs of neurons. Some investigations, including some of ours, indicated that activation of neuronal CB1 receptors induces neuroprotection in ischemic brain injury (Knowles et al., 2016; Du et al., 2010; Sun et al., 2016), and our another study showed that activation of neuronal mitochondrial CB1 receptor contributed to the cannabinoid CB1 receptor agonist ACEA-induced neuroprotection against brain ischemia (Ma et al., 2015). Therefore, in this study, we investigated how the CB1 receptor agonist ACEA gives birth to neuroprotection in neuronal mitochondria against brain ischemia.

Mitochondrial permeability transition pore (MPTP) is comprised of three components, including the voltage-dependent anion channel (VDAC) locating at the outer cell membrane, the adenine nucleotide translocase (ANT) in the inner mitochondrial membrane, and cyclophilin D (Sun et al., 2012). MPTP opening follows brain I/R injury, which can result in mitochondrial inner membrane potential collapse, destruction of respiratory chain, mitochondrial swelling and releases of Cyto C and AIF (Ye et al., 2012). After the MPTP opening, Cyto C in mitochondria can be released into cytosol, and mitochondrial AIF can be released into cell nucleus. Therefore, by testing the expressions of Cyto C in cytosol and AIF in cell nucleus, we can know the MPTP opening level. Moreover, we evaluated the Cyto C and AIF expressions in mitochondria and the whole neuronal cells in vitro. We found that the levels of inner mitochondrial Cyto C and AIF reduced at 24 h after the OGD injury. In contrast, the levels of Cyto C in cytosol and AIF in cell nucleus increased after the OGD injury.

However, the intracellular Cyto C and AIF expressions were not changed. These findings showed that cannabinoid CB1 receptor agonist ACEA inhibited MPTP opening in the ODG-treated neurons. In this study, we used MPTP opener atractyloside (Atr) to observe whether the Atr can abolish the CB1 agonist ACEA-induced neuroprotective effects. Surprisingly, we found that Atr significantly reversed the ACEA-induced protections against brain ischemia in vivo and against OGD injury in vitro. And the MPTP inhibitor cyclosporin A (CsA) did not induce marked effects on the protections caused by ACEA. Because Atr is the ligand of ANT, which can reverse the CsA-induced effects on the MPTP opening by inhibiting the conformational change in mitochondria (Basso et al., 2008). In addition, some studies indicated that MPTP inhibitor CsA can bring about neuroprotective effects against brain ischemia (Li et al., 2009; Yamaguchi et al., 2006; Yu et al., 2004). These evidences supported that the ACEA induces neuroprotection against brain ischemia via inhibiting the MPTP opening. Mitochondrial swelling is one of the initial changes after brain ischemia (Sun et al., 2014; Panickar et al., 2015).

In this study, we tested the mitochondrial swelling level in the OGD-treated neurons, and the result can reflect the mitochondrial membrane permeabilization. We found that OGD increased the mitochondrial swelling level and enlarged the mitochondrial morphology. And the CB1 receptor agonist ACEA-induced inhibition of mitochondrial swelling and enlargement were significantly abolished by the MPTP opener, but not by the MPTP inhibitor. As the mitochondrial swelling can increase the MPTP opening and Cyto C release from mitochondrial into cytosol, and the released Cyto C can form the apoptosome in cytosol (Song et al., 2015), our findings also indicated that inhibition of MPTP opening may be involved in the ACEA-induced neuroprotection. After the brain I/R injury, except for inflammation, excitatory toxicity and apoptosis, oxidative stress plays a vital role in the secondary brain injury (Won et al., 2015). Large amount of reactive oxygen species (ROS) could be generated after the reperfusion, and that can damage the neuronal membrane and mitochondria by producing oxidants and consuming the intracellular anti-oxidants (Li et al., 2015).

In this study, we found that OGD plus re-oxygenation increased the mitochondrial superoxide level and reduced the mitochondrial SOD2 expression. Superoxide dismutase (SOD) consists of cytosolic SOD1, mitochondrial SOD2 and extracellular SOD3. High level of SOD2 can reduce the oxidative injury to mitochondria (Sun et al., 2016). In this study, we observed that OGD plus re-oxygenation increased the mitochondrial superoxide level and reduced the SOD2 in mitochondria, and the CB1 receptor agonist ACEA decreased the superoxide and ameliorated SOD2 expression in mitochondria. The ACEA-induced effects on mitochondrial superoxide and SOD2 expression were reversed by MPTP opener Atr, but not by the MPTP inhibitor CsA. Similarly, one study by Sun et al. showed that cannabinoid CB1 receptor activation reduced brain ischemic injury by increasing SOD2 expression in ischemic brain injury (Sun et al., 2016). And another study of us indicated that endocannabinoid agonist AEA reduced hydrogen peroxide-induced oxidative injury in mouse hippocampal neurons via CB1 receptor (Jia et al., 2014). Mitochondria are the “energy factory” of cells.

After the ischemia, mitochondrial functions in the ischemic brain tissue could be inhibited, including reductions of mitochondrial membrane potential (MMP) and mitochondrial complex activities (Chen et al., 2017; Sanderson et al., 2013). Reductions of neuronal mitochondrial functions may lead to neuronal injury and even death. In this study, we investigated the ACEA-induced effects on mitochondrial functions by testing the MMP and mitochondrial complex activities in OGD-treated neurons.
